Inspiration
Traditional pharmaceutical manufacturing often relies on broad, standardized dosage strengths (e.g., 500 mg, 650 mg), which lack the flexibility needed for personalized medicine. We were inspired to bridge the gap between micro-dosing and mass-scale production—enabling exact titrations for sensitive, pediatric, or critical patients without compromising product quality, assay uniformity, or bioequivalence.
What it does
Project Hriddho 03 establishes a systematic matrix framework that classifies every single dosage increment—from a 1 mg micro-dose up to a 1500 mg maximum dose—as a distinct, standardized proprietary therapeutic product.
By applying a dynamic micro-excipient mass balancing formula ($M_{\text{total}} = M_{\text{API}}(d) + M_{\text{excipient}}(d) = \text{Constant}$), the total tablet weight is adjusted so that every single product variant, regardless of active ingredient dose, yields identical quality control analytical outcomes (e.g., HPLC assay purity within 98.0%–102.0%) and standardized dissolution profiles.
How we built it
- Mathematical & Chemical Matrix Modeling: Developed theoretical formulation balance equations using Paracetamol ($C_8H_9NO_2$) as a baseline active model.
- Excipient Balancing System: Integrated microcrystalline cellulose, lactose monohydrate, and magnesium stearate as binder/filler buffers to normalize overall mass.
- Analytical Standardization: Standardized Reverse-Phase High-Performance Liquid Chromatography (RP-HPLC) protocols (C18 column, 243 nm UV detection) to validate assay consistency across micro and macro dose spectrums.
- Spectrum Mapping: Designed an automated product matrix assigning proprietary codes (e.g.,
Hriddho-PCM-0001toHriddho-PCM-1500) with precise API/excipient weight ratios.
Challenges we faced
- Assay Consistency at Ultra-Low Doses: Ensuring that a 1 mg dose tablet yields the exact same percentage purity assay and HPLC peak retention time as a 1500 mg tablet required rigorous dilution factor adjustments and excipient compatibility testing.
- Mass Balancing: Designing an adaptable filler/binder matrix that maintains structural tablet integrity across extreme API mass ratios.
What we learned
- How micro-excipient mass normalization can stabilize bioequivalence standards across continuous dose ranges.
- Advanced RP-HPLC analytical assay methodologies for low-dose micro-encapsulations.
- The logistical and regulatory considerations required for mass-scale standardization of custom titration pharmaceuticals.
What's next for Hriddho 03
- In Vitro & In Vivo Dissolution Profiling: Conducting real-time dissolution kinetic studies to confirm bioequivalence across all matrix tiers.
- AI-Powered Formulation Engine: Developing automated software that dynamically calculates exact excipient adjustments for any active pharmaceutical ingredient (API).
- Scale-Up & Industry Integration: Collaborating with pharmaceutical manufacturers to test pilot batch runs and patent the custom-titration matrix framework.
