Everyone in the world noticed that the severe COVID 19 infected patients are dying due to multiple organ failure. This organ failure happens to COVID 19 patients due to Cytokine storm. The uncontrolled release of inflammatory cytokines in response to pathogens is known as cytokine storm. Researchers reported that high levels of various cytokines and pro-inflammatory factors such as CXCL10, CCL7 and IL-1 receptor antagonist, interleukin (IL)-6, IL-2R, IL-8, tumor necrosis factor-α, and granulocyte-colony stimulating factor were observed in the Covid 19 infected patients. Targeting individual cytokines does not seem to increase the viral infection rates or stimulate a more severe course of viral infection.(4) Based on these findings we decided to investigate the IL6 and IL-6h inhibiting ability phytochemicals obtained from natural origin such as plant and fungus (eg. Sirolimus, everolimus, Pegylated sirolimus, sirolimus formulated in the form of inclusive complex ..etc). At this moment we focused on sirolimus because it is approved by FDA as an immune suppressant in organ replacement therapy. The mode of action of sirolimus is to bind the cytosolic protein FK-binding protein 12 and inhibit cytokines. Further modifying its nature by PEGylation or converting as an inclusive complex it prolongs and improves its activity inside the human body. As these drugs are already available in the market and upon the successful identification and developing suitable delivery systems for this drug, candidates provide immediate outcome by saving the life of a greater number of severely infected COVID 19 patients.
