- Nicotine dehydrogenase (NDH) - This carefully selected bioreceptor demonstrates exceptional specificity for nicotine, ensuring accurate detection without interference from other compounds.
- Gold electrode - Gold Electrode: The gold electrode serves as the substrate for immobilizing NDH and facilitating electron transfer, playing a pivotal role in the signal transduction process.
- Self-assembled monolayer (SAM) of thiolated alkanethiol - Thiolated alkanethiol SAM provides a stable and customizable interface for anchoring NDH, preserving its activity and enhancing the biosensor's performance.
- Phosphate-buffered saline (PBS) - This solution serves as the medium for facilitating the diffusion of nicotine and the subsequent interaction with the immobilized NDH.

Assembly Process:
Cleaning the Gold Electrode
- Submerge the gold electrode in piranha solution (3 parts sulfuric acid to 1 part hydrogen peroxide) for cleaning.
- Allow it to soak for a few minutes, ensuring all surfaces are exposed to the solution.
- Rinse the electrode thoroughly with deionized water to remove any residue from the cleaning solution.
SAM Application
- Dry the cleaned gold electrode using a lint-free cloth or nitrogen gas.
- Apply a self-assembled monolayer (SAM) of thiolated alkanethiol to the electrode surface.
- Ensure even coverage and adherence of the SAM to the gold electrode.
Bioreceptor Immobilization
- Immobilize Nicotine dehydrogenase (NDH) on the SAM-modified electrode surface.
- Apply a controlled amount of NDH solution onto the SAM-coated electrode.
- Allow for sufficient time for NDH to bind and adhere to the SAM.
Connecting to Potentiostat
- Connect the modified gold electrode to a potentiostat using appropriate cables and connectors.
- Ensure secure and proper electrical connections between the electrode and the potentiostat.
Solution Preparation
- Prepare a solution of phosphate-buffered saline (PBS) in a suitable container.
Setting up the Electrode
- Submerge the NDH-modified gold electrode into the PBS solution.
- Ensure that the electrode is fully immersed and positioned securely.
Nicotine Introduction
- Add a controlled amount of nicotine to the PBS solution.
